Apolipoprotein E gene (APOE) is a cholesterol-carrying protein involved in lipid metabolism, neuronal growth, synaptic maturation, and cellular repair. The composition of APOE, which has three polymorphic alleles (E2, E3, and E4), is a major risk factor Alzheimer’s disease (AD). The presence of two APOE E4 alleles is the highest risk factor for developing AD as well as non-AD age related cognitive decline. While the contribution of APOE E4 in AD pathogenesis is thought to involve altered lipid transport and metabolism in neurons and glia, additional mechanisms underlying its contribution to AD are still being elucidated.
iCell GABANeurons AD APOE E4/E4 were developed as cellular model to investigate the mechanisms underlying ApoE4/E4 risk in developing AD as well as potential therapeutic treatments. The APOE E4/E4 cell line was engineered from the isogenic parental APOE E3/E3 induced pluripotent stem cell line, followed by differentiation into cortical neurons that exhibit a majority GABAergic phenotype.
